Carbomer is a white loose powder with strong hygroscopicity. Because its molecular structure contains 52% to 68% acid groups, it has a certain acidity. The pH value of carbomer 1% water dispersions is 2.5 to 3. It can neutralize the carboxyl groups of the molecules and form gels by neutralizing basic substances. What factors do we need to pay attention to when preparing carbomer gel
1. Selection of gelling agent
When preparing water soluble gel, the carbomer concentration is generally 0.5% to 2% according to the viscosity of the gel. When the emulsion gel was prepared, the concentration was 1%. Considering the transparency and thickening rate of gelatin, the effect of carbomer 940 is more obvious.
2. Selection of neutralizer
Carbomer is a kind of synthetic polyacrylic acid compound, which contains a lot of free carboxyl group and has certain acidity. When the pH value is higher than 4, the carboxyl group begins to dissociate, the polymer dissolves and the viscosity increases; When the pH value is 8, the dissociation is basically completed and the viscosity is the highest. Therefore, neutralizer (pH regulator) is needed to prepare the gel to adjust the viscosity of the preparation. Inorganic alkali neutralizers include sodium hydroxide and potassium hydroxide, while organic alkali neutralizers include triethanolamine, ethylenediamine, laurylamine and stearic acid amine, and triethanolamine is commonly used. When carbomer is used as emulsifier, inorganic alkali neutralizer and organic alkali neutralizer can be used for double neutralization. Carbomer neutralized by inorganic alkali can be dissolved in water phase, and that neutralized by organic alkali can be dissolved in oil phase. The emulsion has excellent chemical and mechanical stability. Generally, 35g triethanolamine or 400mg sodium hydroxide is used for 1g carbomer.
3. Selection of moisturizer
Because the gel itself is not properly stored or packaged, it has appeared to be dry hardening, forming a dry gelatin and not easy to spread. Moisturizing agent is added to maintain the water content of the gel and maintain the stability of the dosage form. Commonly used humectants are glycerin, propylene glycol and other polyols.
4. Selection of stabilizer
The presence of metal ions and strong electrolytes will reduce the viscosity of the gel. At this time, stabilizers can be added to the formulation. Sodium pyrosulfite, citric acid, tartaric acid and EDTA are commonly used. Generally, 0.01% ethylenediamine tetra acetic acid two sodium (EDTA-2Na) solution is used as the metal complexing agent.
5. Selection of antimicrobial agents
Gel is a semisolid preparation, susceptible to microbial infection, especially in humid environment, adding proper preservatives can play a certain bactericidal or bacteriostatic effect. Appropriate preservatives should be selected according to different environments, such as sodium benzoate, p-hydroxybenzoate (Nipagin) and ethanol. It is worth noting that the commonly used quaternary amine salts can not be compatible with Carbomer, because carbomer is sensitive to salt and easy to cause precipitation.
