PIPES is a member of the ethanesulfonic acid buffer series, and its Chinese name is 1-4-piperazine diethanesulfonic acid. It is one of the ethanesulfonic acid series buffer reagents developed by Good and others in 1960. Important features such as medium pKa, maximum water solubility and minimum solubility in all other solvents, minimum salt effects, minimum pKa change with temperature, (PIPES is poorly soluble in water) chemically and enzymatically stable, minimum absorption in visible or UV spectra Features such as range, ease of synthesis, etc.
The effective buffer range of PIPES is pH 6.1-7.5, pKa=6.8 (25℃). Its pKa is close to physiological pH, suitable for cell culture. In addition, in animal and plant histochemistry, PIPES can also act as a buffering agent to minimize lipid damage and experimental artifacts caused by glutaraldehyde fixation. It is worth noting that PIPES is not suitable for application to oxidized reductants due to its ability to form free radicals.
PIPES can be used in in vitro cell culture medium, and in the study of lactate oxidase method for the determination of lactate in plasma, PIPES was found to be stable for 172 hours at 37°C. For immunofluorescence labeling, 4% paraformaldehyde in 50 mmol/L PIPES buffer was used for fixation.
