Carbomer is used as a sustained-release agent of chemical drugs
Carbomer is widely used as a chemical sustained-release agent, including oral and external use. It is in the stage of rapid development and research in China. In this paper, a kind of toxin carbomer gel is used to illustrate the application characteristics, drug release mechanism, preparation process, release factors and practical application of carbomer sustained release gel. Toxoid, i.e. force reducing toxin and denatured toxin, refers to some toxins that lose their original toxicity and still retain their immunogenicity after denaturation or chemical modification. Sustained release agent of carbomer for tetanus toxoid
Bacterial exotoxin is treated with 0.3% ~ 0.4% formaldehyde to remove its toxicity and can be made into toxoid. Tetanus (Clostridium tetanus) toxoid is one of them. Study on its release in carbomer (flocculent precipitation method): dissolve 25lf / ml tetanus toxoid in phosphoric acid buffer and distilled water respectively, then add the same amount of Carbopol with three concentrations of 0.25%, 0.125% and 0.625%, fully mix, separate Carbopol with NaCl precipitation method, and then take the supernatant and Carbopol precipitation for flocculent reaction respectively.
The results showed that the content of tetanus toxoid in the supernatant was very high and showed typical flocculent precipitation, while the Carbopol precipitation did not contain any tetanus toxoid, and the flocculent reaction was negative. This study preliminarily established the biological detection method of the product in carboper gel and the conditions for its dissolution in carboper. It was determined that the concentration of Carbopol in this release study should not be higher than 0.25%.
