PIPES, fully known as piperazine-1,4-diethylsulfonic acid, can maintain a basically constant pH value of the solution within a certain range. When a certain amount of acid and alkali is added to some solutions, it can hinder the change of pH of the solution, which is called buffer biological buffer. Such solutions are called buffer solutions. Pipes is an important member of good's buffer. Good's buffer is a kind of buffer system dedicated to life science research, which does not participate in and interfere with biochemical processes.

The pH buffer range of pipes is 6.1-7.5, insoluble in water and soluble in NaOH aqueous solution. Pipes is different from buffers containing di (2-hydroxyethyl) amino groups (such as bis Tris, bicine). It can not form stable complexes with most metal ions. It is suitable for buffers in solution systems containing metal ions. According to previous studies, PIPES can be used to purify tubulin by using cellulose phosphate chromatography, and to purify recombinant GTP binding protein ARF1 and ARF2 by gel filtration as a buffer to crystallize ketoenzyme from E. coli. Because pipes can form free radicals, it is not suitable for redox system. In cation exchange chromatography, low concentration pipes buffer should be used because pipes has relatively large ionic strength and its pKa value is concentration dependent.
Preparation method of pipes solutions with different pH values (about 1-2l)
1. Prepare 0.1M solution (a)
