Details
pipes , Chinese name is 1-4-piperazine diethylsulfonic acid. It is an amphoteric buffer with pH buffer range of 6.1-75. It is insoluble in water and soluble in NaOH aqueous solution. It is at pH 7 It has good buffer capacity in the range of 2-74. When a certain amount of acid and alkali is added to some solutions, it can hinder the change of pH of the solution, which is called buffering. Such a solution is called buffering solution.
Application of pipes buffer:
Pipes buffer is widely used in cell culture, chromatography, cosmetics, diagnostic testing, protein purification and electron microscopy. Pipes buffer is similar to MES buffer and mops buffer, but it lacks the ability to form chelates with most metal ions, so it is recommended to be used as non coordination buffer in metal ion solution.
Advantages of pipes buffer:
Recently, it has been learned from professional literature that in the glutaraldehyde solution used in the ultrastructural observation experiments of various tissues such as human skin, human hypertrophic scar and mollusc nerve tissue, the difference between adding NaOH pipes buffer and pipes buffer is compared. The results showed that the addition of NaOH pipes buffer could help researchers observe more details between organelles and tissues, and the consistency of cytoplasm was more uniform. If NaOH pipes buffer is added, microfilaments and microtubules can be observed more generally. In addition, it was proved that pipesbuffer was comparable to living cells in culture.
Pipes is different from buffers containing di (2-hydroxyethyl) amino groups (such as bis Tris, bicine). It can not form stable complexes with most metal ions. It is suitable for buffers in solution systems containing metal ions. According to previous studies, PIPES can be used to purify tubulin by using cellulose phosphate chromatography, and to purify recombinant GTP binding protein ARF1 and ARF2 by gel filtration as a buffer to crystallize ketoenzyme from E. coli. In addition, pipes can form free radicals, so it is not suitable for redox system. In cation exchange chromatography, low concentration pipes buffer should be used because pipes has relatively large ionic strength and its pKa value is concentration dependent.
