The biological buffer Taps can be used as a buffer for nucleic acid DNA or RNA in capillary zone electrophoresis. Capillary zone electrophoresis is different from electrophoresis in electrophoresis tank. Electrophoresis in electrophoretic tank usually requires separation of DNA or nucleic acid from agarose gel or other gel gels, and separation of various particles can be carried out directly in capillary tubes.

When the pH value of the quartz capillary column used for thin tube electrophoresis is > 3, its inner surface is negatively charged, and an electric double layer is formed when it is in contact with the buffer. Under the action of high voltage electric field, the buffer on one side of the electric double layer moves to the negative direction due to positive charge, thus forming electroosmotic flow.
At the same time, in the buffer solution, the charged particles move to the opposite direction of the charge polarity at different speeds under the action of electric field to form electrophoresis.
The migration velocity of charged particles in capillary buffer is equal to the vector sum of electrophoresis and electroosmosis. Various particles are separated due to different migration speeds caused by different charge, mass, volume and shape.
The stationary phase of HPLC can also be filled into the capillary or coated on the inner wall of the capillary to form capillary electrochromatography. This mode has the separation mechanism of electrophoresis and liquid chromatography.
