【Use】 Trimethylamine methane is widely used in acute metabolic and respiratory acidemia. It is an alkaline buffer, which has a good buffer effect on metabolic acidosis and enzyme activity reaction, which can cause hypoglycemia, hypotension, nausea and vomiting, and also inhibit respiration and even stop the breath. Alkalinity can be caused by excessive intake or renal insufficiency. Tris (trimethylamine) is often used as biological buffer, and is often prepared to pH 6.8, 7.4, 8.0 and 8.8. The structure of the system is characterized by a large change of pH with temperature. Tris (trimethylamino methane) is widely used in the preparation of buffer in biochemical and molecular biology experiments. Tris (trimethylamine) is weak base, and its PKA is 8.1 at 25 ℃ at room temperature; According to the buffer theory, the effective buffer range of Tris buffer is between pH7.0 and 9.2. The pH of Tris alkali aqueous solution is about 10.5. Hydrochloric acid is generally added to adjust the pH value to the required value, and the buffer solution of the pH value can be obtained. However, the effect of temperature on the pKa of Tris should be noted. Because Tris buffer is a weak alkaline solution, DNA will be depolymerized in such a solution, thus improving its solubility. EDTA is often added to Tris hydrochloric acid buffer to make "Te buffer", which is used for DNA stability and storage. If the acid solution adjusting the pH value is replaced with acetic acid, the "Tae buffer" (tris/acetate/edta) is obtained, and the "tbe buffer" is obtained by replacing with boric acid. These two buffers are usually used in nucleic acid electrophoresis experiments. TAE and tbe, which are made from Tris, are the most commonly used reagents in DNA electrophoresis. Te (ph8.0) is mainly used to dissolve DNA( Te is the joint name of Tris and EDTA.) 1mtris hcl6.8 and 1.5mtris-hcl8.8 are the most commonly used reagents in SDS PAGE.
