Details

Dipotassium ethylenediaminetetraacetate, or EDTA dipotassium for short, is a very important anticoagulant in vacuum blood collection. It is used more frequently than anticoagulants such as heparin, sodium citrate and potassium oxalate. Therefore, it is very helpful to understand some details of its use.

Application type of EDTA dipotassium

The anticoagulants added by different types of anticoagulant tubes are naturally different. EDTA dipotassium is usually added to the vacuum blood collection vessel for blood routine test, which is generally the tube with purple safety cap. In addition, in nucleic acid detection tubes, potassium EDTA is also generally used as blood anticoagulant. First, EDTA can complex calcium ions in the blood and inhibit the coagulation process; Secondly, EDTA can also complex magnesium ions, which can inhibit nucleases that need the presence of magnesium ions to play a role, and prevent nucleic acids from being degraded by nucleases.

Amount of potassium EDTA

The dosage of dipotassium EDTA in blood collection vessels is determined according to the blood collection volume. According to the requirements of appendix NC of yy0314-2007 standard, the content of dipotassium EDTA or Tripotassium EDTA in each ml of blood is 1.5-2.2mg, and the intermediate value of 1.8mg/ml is taken when preparing reagent. It should be noted that the dosage of heparin anticoagulant is 20iu per ml of blood, one is measured by weight and the other is measured by titer unit. The two cannot be confused.

Preparation of potassium EDTA solution

EDTA dipotassium is a salt that is easily soluble in water. When it is prepared, it can be directly dissolved in water. However, there will be a small amount of dilution when EDTA dipotassium or Tripotassium anticoagulant is mixed with blood samples. In order to control the effect of dilution, the concentration of EDTA solution should not be too low. Usually, 20 μ l of dipotassium solution is added to 2ml or 3ml blood collection. When the amount of dipotassium needs to be increased, the method of increasing the concentration of dipotassium takes precedence over increasing the spraying amount. However, the concentration of potassium dioxide should not be too high. Too high concentration may reduce the solubility when the temperature decreases, resulting in the precipitation of potassium EDTA crystal, and too high concentration of potassium ion may cause the risk of hemolysis of blood cells.

The anticoagulant principles of potassium EDTA and potassium EDTA are the same. After preparing the solution, only the concentration of EDTA acid radical ion and potassium ion are different, there are slight differences in pH, and there are certain differences in specific test items.
