Compared with traditional techniques such as radioimmunoassay (RIA) and enzyme-linked immunosorbent assay (ELISA), chemiluminescence immunoassay (CLIA) is a new technology based on chemiluminescence (CL) reaction, which has attracted much attention in recent ten years. Due to its wide dynamic range and high sensitivity, this technology has quickly become the main method of clinical immunological analysis. This method is widely used in the detection of tumors, infectious diseases, metabolic diseases and pregnancy.
There are three chemiluminescence immunoassay methods:
I Direct chemiluminescence immunoassay
Commonly used markers: isoluminol and acridine ester. These markers can produce special luminous groups and directly participate in the reaction in the analysis process.
1. Direct labeling chemiluminescence immunoassay with acridine ester: after immune reaction with the corresponding antigen (antibody) in the sample to be tested, a solid-phase coated antibody acridine ester labeled antibody complex is formed. At this time, only oxidant (H2O2) and NaOH are added to form an alkaline environment, and acridine ester decomposes and emits light without catalyst.
2. Chemiluminescence immunoassay of luminol labeling: the luminescence of luminol is oxidation reaction luminescence. In alkaline solution, luminol can be oxidized and luminescent by many oxidants, among which H2O2 is the most commonly used. Due to the slow luminescence reaction, some enzymes or inorganic catalysts need to be added. The enzymes are mainly horseradish peroxidase (HRP), and the inorganic ones include O3, halogen, Fe3, Cu2, CO2 and their complexes.
II Indirect luminescence immunoassay
The commonly used markers are horseradish peroxidase (HRP, the commonly used substrate is luminol or its derivatives, such as isoluminol) and alkaline phosphatase (ALP, the commonly used substrate is 1,2-dioxane derivative, amppd). These markers participate in the luminescence reaction as a catalyst or energy transfer receptor of the luminescence reaction.
III Electrochemiluminescence immunoassay
The common marker is tripyridine ruthenium (the common substrate is tripropylamine).
